Catatan | ABSTRAK :Rapid diagnosis played an important role during the outbreak of anthrax. Diagnosis of anthrax in Indonesia is now made by using conventional methods. Direct fluorescent antibody assays (DFA) have been developed to rapidly identify B. anthracis cultures and to directly detect B. anthracis in infected clinical specimens. The utilization of Fluorescent labelled antibodies specific to the B. anthracis cell wall polysaccharide antigen (CW-DFA) for diagnosing anthrax has been recommended by Center for Disease Control and Prevention, USA. The component used in DFA assay is polysaccharide-peptidoglycan complex of B. anthracis cell wall, which was prepared from B. anthracis culture by cell lysis and guanidine extraction. The purified polysaccharide-peptidoglycan complex was mixed with Freunds complete/incomplete adjuvant and then used for immunizing goats. The IgG was then labeled with Fluorecent Iso Thiocyanate, ready for use as conjugates in DFA Assay. The CW-DFA developed are specific to the B. anthracis cell wall antigens. The assay was evaluated for rapid identification of B. anthracis, B. anthracis isolates and non B. anthracis isolates were tested by the CW-DFA assay. B. cereus, B. subtilis, and other Bacillus spp were negative, while B anthracis gave positive results. The CW DFA assay is a specific rapid confirmatory test for B. anthracis in cultures. / Diagnosa yang cepat mempunyai peranan penting dalam kejadian kasus antraks. Diagnosis penyakit antraks di Indonesia sampai saat ini masih menggunakan cara konvensionil. Direct fluorescent Antibody (DFA) assay telah dikembangkan untuk secara cepat dapt mengidentifikasi kultur B. anthracis dan secara langsung mendeteksi B. anthracis dalam spesimen yang mengandung B anthracis. Penggunaan antibodi yang dilabel dengan fluorescein specifik terhadap antigen polisakarida dinding sel B. anthracis. (CW-DFA) dalam diagnosis penyakit antraks telah direkomendasikan oleh Center for Disease Control and Prevention, USA. Komponen yang digunakan dalam DFA assay adalah polisakarida peptidoglikan dari dinding sel B. anthracis yang dibuat dari kultur B anthracis dengan cara melakukan lisis terhadap sel dan ekstraksi guanidin. Polisakarida-peptidoglikan kemudian dicampurkan dengan adjuvan Freunds complete/incomplete untuk imunisasi kambing. Serum hiperium kemudian dikoleksi dan dimurnikan untuk memperoleh IgG anti dinding sel (kompleks polisakarida-peptidoglikan) B.anthracis. IgG yang diperoleh kemudian dilabel dengan Fluorecent Iso Thiocyanate untuk dapat digunakan sebagai konjugat dalam uji DFA. Ternyata CW-DFA yang dikembangkan spesifik untuk antigen dan bukan isolate B.anthracis diuji dengan uji CW-DFA ini. B.cereus, B. subtilis, dan Bacillus spp lainnya memberikan hasil negatif, sedangkan B anthracis memberikan hasil positif. Uji CW_DFA adalah uji yang spesifik dan cepat untuk konfirmasi kultur B. anthracis. |
