The preservation method used should minimize loss of viability during processing and storage so that after preservation, cultures will survive for long periods. Clostridium spp. viability after ex situ conservation by freeze-drying technique with Robertson's cooked meat medium as preservation medium in long storage period at room temperature have been evaluated for achieving the suitable and efficient monitoring in the microbial germ plasma preservation. A total of 376 dried culture samples of 115 Clostridium spp. collection consist of 27 species prepared in vacuum glass ampoules, stored for more than 10 years at room temperature have been evaluated for their viability. Each sample was grown on specific medium and identified for the bacterial species and type. The results showed that 9 of 115 collections, representing 7 different species failed to grow during 12 to 19 years storage. That 7 species were able to grow during 21 to 30 years ex situ conservation by freeze-drying technique with mist. desiccans preservation medium. It is suggested that mist. desiccans preservation medium was better be used in freeze- drying technique to conserve Clostridium spp. germ plasma. aARTVET2341
